The short answer to this is yes.
Only people with a status of "Confirmed" will be sent your emails. When someone clicks the unsubscribe link in an email their status is changed automatically to "Unsubscribed". The system then knows not to send to them. They will remain on your list but because their status is now set to "Unsubscribed" they wont be sent emails.
Note: When someone unsubscribes they are only unsubscribed from the List that the email was sent to. So if you have more than one list and the same email address exists on other lists, they will still remain subscribed to those other lists.
If you require unsubscribing from a list to also unsubscribe them automatically from other lists, then you can configure this in the settings for each list. Edit each list and under Subscriber actions > Actions when unsubscribe, tick the lists they should also be unsubscribed from.
